摘 要:[目的]研究壁虎多糖对培养的H22肝癌细胞体外抑制增殖作用,对荷H22实体型移植瘤小鼠的体内抑制作用,以期寻找有效的抗肿瘤活性成分。[方法]四甲基偶氮唑蓝(MTT)法检测低、中、高剂量组壁虎多糖对H22肝癌细胞体外杀伤作用。采用昆明小鼠H22移植瘤模型观察壁虎多糖高、低剂量组的体内抑瘤活性及荷瘤小鼠胸腺、脾脏指数变化。[结果]体外实验显示:壁虎多糖不同剂量组均可抑制H22细胞的增殖(P<0.05),其抑制率最高为79.1%,远远高于西药48.7%的抑瘤率。然而体内实验中,壁虎多糖各组均无明显抗肿瘤作用,高剂量组的抑瘤率仅为8.8%,各组与模型组比较均无统计学差异(P>0.05)。壁虎多糖高低剂量组与模型组小鼠的胸腺质量、胸腺指数,脾脏重、脾脏指数相比均没有明显差异。而5-氟尿嘧啶(5-Fu)组胸腺质量、胸腺指数,脾脏质量、脾脏指数较模型组和各壁虎组均明显降低(P<0.05)。[结论]体外实验显示壁虎多糖具有良好抑制H22细胞的增殖作用。然而体内实验中,壁虎多糖与模型组比较无统计学差异,壁虎多糖是否是抗肝癌有效成分有待进一步研究。[Objective] To investigate the in vitro inhibitory effect of Gekko polysaccharide on the proliferation of cultured H22 cell of liver cancer in mice. [Methods] MTT assay was applied to evaluate the killing effect of Gekko polysaccharide on cultured H22 hepatoma cells in vitro. The anti-tumor effect of Gekko polysaccharide was observed in H22 tumor bearing mice. [Results] The results showed each dose of Gekko polysaccharide group had effective anti -proliferation effect on the H22 cells in vitro. The highest inhibition ratio was 79.1% ,far more higher than 5-FU group, in which inhibition ratio was 48.7%. This result was consistent with Wu. The inhibitory rate of high dose of Gekko polysaecharide, however, was only 8.8% in vivo. There was no difference when compared with the control group (P〉 0.05). The influence to the thymus, thymus index, spleen and spleen index of mice with transplanted H22 tumors in each Gekko polysaccharide groups were not different with those in the control group (P〉0.05). In the 5-FU group these indexes were obviously depressed when compared with those in the control group and Gekko polysaccharide groups(P〈0.05). [Conclusion] Gekko polysaccharide has good anti-tumor effect in vitro. However, the inhibition rate of Gekko polysaccharide was low in vivo. Experiments in vitro and in vivo should be combined to make experimental results much more persuasive. Whether Gekko polysaccharide of gecko was the anti-tumor active ingredients remains further explored.
