摘  要:目的探讨缺氧诱导因子(HIF-1α)基因治疗家兔肢体缺血模型的疗效。方法在家兔缺血模型的肢体内注射携带/HIF-1α基因的重组腺病毒AdHIF-1α,检测HIF-1α基因在缺血肌组织中表达情况,并观察其生物学特点。实验以携带GFP基因的重组腺病毒AdGFP为对照。结果免疫组化染色均检测到HIF-1α的表达。转AdHIF-1α基因治疗组毛细血管计数及小动脉计数均高于AdGFP转导的对照组(P<0.05)。动脉造影证明基因治疗组促进缺血肢体侧支循环建立更为有效。结论家兔缺血肢体内注射AdHIF-1α能够获得外源基因蛋白质的有效表达,能有效的促进血管生成及侧支循环建立。Objective To investigate the efficacy of HIF- 1α gene transfer in a ischemia model in hindlimb of rabbits. Methods To establish an experimental animal model of limb ischemia. Constructed AdHIF-1α expression plasmid was directly injected into the ischemia hindlimb of rabbits. Immtmotfistochemistry, capillary density and arteriole density were used to detect gene expression and biological effect. Artery angiograpby was done to evaluate collateral circulation of the occluded artery. Results High levels of HIF- 1α protein expression were detected. There was a significant increase in the number of capillaries and arterioles as compared with a control group ( P 〈 0.05). Capillary density and arteriole density of the therapy group were higher than those of the control group. Conclusion Direct injection of AdHIF-1α can transfect the ischemia muscle and express HIF-1α protein. Gene transfer of HIF-1α can increase capillary and arteriole number and enhance collateral circulation of the occluded artery more effectively.
