摘  要:采用荧光光谱法、电子吸收光谱法及循环伏安法研究了配合物LCu(Ⅱ)(L=二甲醇-二硝酸-二[1,3-二(苯并咪唑-2)丙烷])与DNA的相互作用.荧光光谱法研究显示,不同浓度的配合物与DNA作用有不同的结合常数及n值,表明配合物与DNA之间呈混合型作用模式.加入DNA后电子吸收光谱产生减色效应及红移现象,结合常数K为2.42×103L/mol,表明铜配合物与DNA发生了弱插入作用.循环伏安法研究显示,加入DNA后峰电位明显负移,峰电流下降,表明发生静电作用;而DNA浓度较低(约5.0×10-4mol/L)时,随着DNA浓度的增大峰电位又有相对正移现象,表明可能同时存在弱插入作用.综上所述,配合物LCu(Ⅱ)与DNA发生了弱插入作用和静电作用.The binding of DNA with the LCu ( Ⅱ) (L = bis [ 1,3-bis (benzimidazol-2-yl) propane-k^2 N, N'] dinitrate methanol) has been investigated by fluorescene,absorption spectroscopy and cyclic voltammetry methods. The fluorescence experiments show a mixed effect between DNA and the copper complex. Absorption spectra show that the absorption peaks exhibit hypochromicity and red shift when DNA is added. The binding constant is 2.42× 10^3 L/mol by the absorption spectra. These results indicate that the copper(Ⅱ) complex can intercalate weakly into the base-pairs of DNA as we calculate it. When DNA was added, the peak potentials in the cyclic voltammogram significantly shift negatively while the peak current decreases. These illuminate there is electrostatic interaction between DNA and the copper complex. But when DNA is in the concentration of 5.0 × 10^-4 mol/L, the positive shift of peak potential with the increasing concentration of DNA means intercalating effect. It can be concluded that the binding of complex to DNA is electrostatic and weakly intercalative.
