摘  要:本试验对可积累聚β-羟基丁酸酯(PHB)的隐藏嗜酸菌(Acidiphilium cryptum)DX1-1菌株的培养条件进行了优化.确定了最佳碳氮源组合(葡萄糖,KNO3),正交试验确定了最优培养条件即碳源浓度为40 g/L、氮源浓度为15 g/L以及初始pH值为3.0,并确定了在最优培养条件下生长曲线和PHB产量随时间的变化曲线.通过核磁共振对该菌胞内PHB的结构进行了分析.此次试验提高了DX1-1菌株PHB的产量,最高达到19.75 g/L.This experiment optimized the culture condition of Acidiphilium cryptum DX1-1, which was capable of producing poly-β-hydroxybutyrate (PHB) . Glucose and KNOa acted as the best carbon and nitrogen substrates for the production of PHB. The L16(4^3) orthogonal experiment improved the yield when 40 g/L glucose as carbon source, 15 g/L KNO3 as nitrogen source and pH 3.0 were adopted. Then the cells growth curve and PHB production curve were measured under the optimum culture condition. It is found that the process of PHB accumulating was a growth-associated process. NMR analysis further supported that the product accumulated by that strain had the typical structure of PHB. The PHB production of strain DX1-1 increased to 19.75 g/L.
