免疫球蛋白阳性细胞在大菱鲆免疫相关组织中的分布

摘　　要：采用单克隆抗体免疫组化标记、间接荧光标记法(IFAT)和流式细胞术(FACS),对大菱鲆免疫相关组织中免疫球蛋白(Ig)阳性细胞进行了检测.免疫组化标记后发现,鳃组织中Ig阳性细胞存在于鳃小片上皮细胞与支持细胞之间的间隙中,后肠Ig阳性细胞存在于固有层和粘膜下层中,外周血血涂片中可见Ig阳性细胞,头肾Ig阳性细胞则无规律地散布于肾组织中.间接荧光标记和流式细胞仪分析免疫相关组织中分离的白细胞,发现大菱鲆不同免疫相关组织中Ig阳性细胞的数量不同,Ig阳性细胞数量分别为:鳃(1.60±0.71)%,后肠(3.91±1.06)%,外周血(28.95±0.39)%,头肾(21.54±0.90)%.结果表明,大菱鲆的免疫相关组织(含粘膜组织)具备一定的完成特异性免疫的细胞基础,且不同组织之间的免疫细胞数量存在显著差异.The methods of immunohistochemistry, indirect fluorescent antibodg Test（IFAT）and fluorescence-activated cell sorter （FACS） were used to determine and measure immunoglobulin（Ig） positive cells in immune-associated tissues of turbots. Results were as follows： Ig-positive cells were located in filament sinusoid between the epithelial cells and the supporting cells of gill, and were located in the tunica propria and the submueosa of hindgut. The Ig-positive cells also appeared in the smear of peripheral blood （PBL） and scattered in interstitial tissue of head kidney. The number of Ig-positive cells was different among tissues in turbot. The percentages of Ig-positive cells respectively were （1.60±0.71）% in gills, （3.91±1.06）% in hindgut, （28.95±0.39）% in PBL, （21.54±0.90）% in kidney. These results showed that immune-associated tissues, including mucosal lymphoid tissues, had the cells to conduct a special immune response. The number of immune cells was significanty different among immune-associated tissues of turbot.
