摘  要:目的探讨应阳流式细胞技术测定眼部常见真菌对抗真菌药物敏感性的可行性。方法用流式细胞术药敏试验(Flow Cytometry Susceptibility Testing,FCST)和标准药敏试验(肉汤稀释法M38-A)法检测1株标准镰刀菌和23株临床镰刀菌对两性霉素B(AMB)的敏感性。FCST法以碘化丙啶(PI)为染料,真菌与各浓度药物孵育3h后检测各管平均荧光强度(MFI),最低抑菌浓度(MIC)定义为相于生长对照管MFI增加50%时的最低药物浓度。结果两种检测方法有8株真菌的MIC完全相同,12株相差1个倍比稀释度,2株相差2个倍比稀释度,在1个稀释度范围内两种检测方法的一致性为83.3%,2个稀释度范围的一致性为91.6%。两组结果无统计学差异(P=0.5685)。结论FCST法检测眼部镰刀菌对抗真菌药物的敏感性所得结果与标准法所得结果有良好的一致性,且FCST法具有快速、敏感、准确的优势,是临床检测丝状菌药物敏感性较具发展潜力的一种新方法。Objective To investigate the possibility of determination of the susceptibility of ocular fusarium isolates to anti--fungal drugs by Flow Cytometry.Methods The sensitivity to amphotericin B (AMB)of one standard strain and 23 clinical isolates of fusarium were examined both with Flow Cytometry Susceptibility test (FCST)and broth microdilution method M38-A.In FCST the mean fluorescence intensity (MFI) of the samples was tested after 3 hours' incubation with various concentrations of the drug by using propidiurn iodide (PI)as a fluorescent probe.The minimal inhibitory concentration (MIC)was defined as the lowest concentration of drug that produced a 50% increase in MFI compared to that of the growth control.Results MIC values showed 83.3 % agreement within one drug dilution between the FCST and broth microdilution method and 91.6 % agreement within two drug dilutions.Paired t test values were not statistically significant(P =0.5685 ).Conclusion The MICs obtained by FCST for all ocular fusarium were in good agreement of the resuits of the broth microdilution method.Flow Cytometry susceptibility test is a rapid, accurate and sensitive method in antifungal mean fluorescence intensity testing of mycelial fungus.
