Effects of Tapping,Mechanical Wounding and Exogenous Jasmonates on the Expression of Genes Encoding for Defense Proteins in the Latex of Hevea brasiliensis

摘  要:为了进一步探明巴西橡胶树乳管的防卫功能,笔者采用RT-PCR和Northern-blot技术分析割胶、机械伤害和外源茉莉酸对胶乳几丁质酶、β-1,3-葡聚糖酶和橡胶素基因表达的影响,结果表明:(1)外源茉莉酸和机械伤害均能上调几丁质酶、β-1,3-葡聚糖酶和橡胶素的基因表达,但机械伤害的效应滞后于外源茉莉酸;(2)几丁质酶、β-1,3-葡聚糖酶和橡胶素的基因在割胶树中的表达明显强于在未开割树中的表达;(3)割胶树胶乳中内源茉莉酸含量显著高于未开割树。There are three defense proteins, i.e., chitinase, β-1,3-glucanase and hevein, in the lutoids of latex from rubber tree. These proteins may be accumulated around the laticifer cut to form a biochemical barrier against pathogenic entrance after being released from lutoids which are usually broken during latex flow. Little is known about the mechanism for the regulation of expression of genes encoding for these proteins. The expression of these genes induced by tapping, mechanical wounding and exogenous jasmonates was analyzed by RT-PCR and Northern blot. The expression of genes encoding for chitinase, β-1,3-glucanase and herein was up-regulated by mechanical wounding and exogenous jasmonates, with the effect of the former being weaker than that of the latter. The expression of these genes is remarkably stronger in trees opened for tapping than in the virgin trees. The level of endogenous jasmonates in the latex is significantly higher in the tapped trees than in the virgin trees. The up-regulation of these genes by mechanical wounding and tapping may be due to the increase in the level of endogenous jasmonates in bark tissues and latex. The increase in the level of endogenous jasmonates in latex caused by tapping may play an important role in enhancing the defense of rubber trees against pathogens.
