香蕉内在拮抗细菌研究Ⅰ——菌株B215的分离及分子鉴定

摘　　要：在香蕉枯萎病(Fusarium oxysporum f.sp. cubense,FOC)重病园区,从生长正常的香蕉假茎内分离获得1株细菌,编号为B215。经对峙培养和孢子萌发抑制测定,B215对FOC和香蕉其他几种主要病原菌的菌丝生长和孢子萌发具有良好抑制效果。对峙培养明显抑制香蕉枯萎病菌菌丝生长,抑菌带宽度0.5～0.6cm。菌株B215培养液滤液使香蕉枯萎病菌孢子和菌丝生长点膨大成球状畸形,原生质外泄,细胞崩溃干瘪,其EC50为4.12%。形态学、生理生化试验显示B215为芽孢杆菌(Bacillus);进一步对该菌株做16SrDNA序列测定与分析,表明其与已报道的枯草芽孢杆菌(Bacillus subtilis strain KL-073)具有100%的同源性,二者所构建的系统发育树上处于同一个分支。将B215鉴定为枯草芽孢杆菌(B.subtilis)。An endophytic strain B215 was obtained from the pseudostem of healthy banana plant in the banana plantation seriously infected with Fusarium wilt （Fusarium oxysporum f.sp.cubense, FOC ）. The antagonistic effects of the strain B215 on FOC and other main pathogens of banana were determined by dual culture and conidial germination method. The dual culture of FOC with B215 resulted in inhibitive zones ranging from 0.5cm to 0.6cm. The conidial germination method showed that the fermentation filtrate solution of B215 significantly decreased the conidial germination of FOC, with the ECso being 4.01%. The cells of the hyphae and spore of FOC became morphologically abnormal when cultured on the medium with fermentation filtrate solution of B215. Morphological and physio-bioehemical identification showed that B215 belonged to Bacillus. 16S rDNA sequencing and analysis of B215 showed the 16S rDNA sequence of B215 shared 100% homology with the published sequence of Bacillus subtilis strain KL-073 from Genbank, and both sequences constituted a branch in phylogenetic tree. Strain B215 was hence identified as Bacillus subtilis.
