摘 要:目的:研究小鼠膜性肾小球肾炎(MGN)脾脏淋巴细胞表面Fas(CD95)的表达及其与MGN发病机制的关系。方法:利用Border改良法建立20只MGN小鼠模型(实验组),以10只正常小鼠作对照(对照组),对其脾脏标本进行Fas抗体免疫组织化学染色分析(Envision法),并对两组小鼠外周血涂片进行淋巴细胞百分率计数。结果:光镜下小鼠肾小球毛细血管壁弥漫增厚,电镜下基底膜弥漫增厚伴有钉突形成及足突融合。实验组小鼠脾脏淋巴细胞表面Fas表达明显低于对照组(P<0.05),而外周血淋巴细胞百分率明显高于对照组(P<0.01)。结论:外周淋巴细胞表面Fas功能障碍或缺陷是MGN发生发展的重要因素之一。Objective To investigate the relationship between the Fas (CD95) expression on lymphocytes in spleen of mice and the pathogenesis of mesangial glomerulonephritis (MGN). Methods The 20 MGN mouse models were established by Border's methods, with 10 normal mice used as control group. The spleen sections were immunohistochemically stained with Fas antibody using Envision method, and the percentage of lymphocytes in peripheral blood of the two groups were counted. Results It was observed by optical microscopy that the glomerular capillary wall was thickened diffusely, while it was shown by electronic microscopy that the basal membrane was thickened diffusely with the formation of rate pegs and the fusion of foot process. The number of Fas-positive lymphocytes in spleen in experimental group was much lower than that in control group (P 〈 0.05), but the percentage of lymphocytes in peripheral blood was much higher than that in control group (P 〈 0.01 ). Conclusion The dysfunction of Fas-positive lymphocytes in periphery may be one of the important factors in the development of MGN.
