基质金属蛋白酶-9及其组织抑制物-1在妊娠期糖尿病患者胎盘组织中的表达及其意义

摘　　要：目的:探讨妊娠期糖尿病患者胎盘组织基质金属蛋白酶-9(MMP9)及其组织抑制物-1(TIMP1) mRNA的表达和意义。方法:收集28例妊娠期糖尿病患者胎盘组织,采用原位杂交法从RNA水平检测其MMP9和TIMP1的表达,同时收集18例正常足月妊娠的胎盘组织进行对照。结果:正常妊娠时MMP9和TIMP1 mRNA在胎盘绒毛中呈强阳性表达,而妊娠期糖尿病患者其胎盘绒毛中MMP9和TIMP1 mRNA表达减弱,差异有显著性(P<0.05),其MMP9/TIMP1比值显著小于正常妊娠时的比值,两者比较差异有显著性(P<0.05)。结论:妊娠期糖尿病患者,尤其合并重度子痫前期的患者,其胎盘绒毛中MMP9和TIMP1 mRNA表达均减少,提示妊娠期糖尿病患者存在着胎盘血管重铸异常,易引起胎盘血流灌注不良。Objective To investigate the mRNA expressions of metrix metalloproteinase-9 （MMP9） and tissue inhibitor of metalloproteinase-1 （TIMP1） in the placenta tissue in patients with gestational diabetes mellitus （GDM） and their significance. Methods The expressions of MMP9 and TIMP1 mRNA in the placenta tissues from 28 patients with GDM and from 18 healthy pregnant women were detected by in situ hybridization. Results There was a significant difference between normal pregnancy and GDM. The expressions of MMP9 and TIMP1 mRNA were highly expressed in the placental villi from healthy pregnant women while were weakly expressed in those from patients with GDM （P 〈 0.05）. The ratio of MMP9/TIMP1 was markedly lower in GDM than in normal pregnancy （P 〈 0.05）. Conclusion Both MMP9 and TIMP1 mRNA expressions are decreased in placental villi in patients with GDM, especially those concurrent wtih preeclamsia, indicating the patients have abnormal placental vascular reconstruction leading to impaired placental blood supply.
