摘  要:本文研究了红细胞生成素(EPO)在体外对巨核细胞成熟及血小板生成的影响。两种纯度的EPO在一定浓度范围内,均能使血小板数呈线性增加,但巨核细胞数保持不变。EPO-Ⅰ可使2~8倍体(N)巨核细胞减少,16~64N细胞增多,同时Ⅰ期巨核细胞比例减少,Ⅳ期细胞比例增多。两种EPO以等单位或最大效应(增加血小板数的作用)相比,EPO-Ⅰ的作用强于EPO-Ⅱ。实验表明EPO使体外血小板数升高的作用是由于促进了巨核细胞DNA合成及胞浆成熟。Liquid cultures of mouse bone marrow cells were used to study the effect of two kinds of erythropoietin (EPO, from the urine of aplastic anemia patient) on the maturation of megakaryocytes and platelet release. Both kinds of EPO increased the platelet count (dose dependent) while the megakaryocyte number remained unchanged. Addition of EPO- Ⅰ(specific activity 10U/mg) to cultures resulted in an increase of 16' N -64N cells in contrast with a decrease of 2-8N cells (P<0.01) . Simultaneously, stage Ⅰ cell dropped slightly but significantly and stage Ⅳ cell increased. As compared with EPO-Ⅱ (specific activity 60U/mg) on the basis of either equal units or highest effect (platelet count) , EPO-Ⅰ showed a stronger effect on megakaryocyte maturation and platelet formation. For further investigation, the purer EPO (such as r-EPO) is needed.
