摘 要:目的考察青龙衣多糖对S180小鼠红细胞Ca2+,Mg2+-ATP酶活性及[Ca2+]i的影响。方法青龙衣多糖对S180小鼠ip给药7 d,比色法结合试剂盒测定S180小鼠红细胞膜Ca2+,Mg2+-ATP酶的活性,采用激光共聚焦显微镜结合Fluo-3/AM荧光探针测定红细胞内[Ca2+]i。结果青龙衣多糖能提高S180小鼠红细胞膜Ca2+,Mg2+-ATP酶活性(P<0.05);降低S180小鼠红细胞[Ca2+]i(P<0.05),并且青龙衣多糖的作用呈剂量依赖性,以高剂量组的作用最佳(P<0.01)。结论青龙衣多糖通过提高S180小鼠红细胞膜Ca2+,Mg2+-ATP酶的活性,排除离子跨膜转运障碍,稳定其能量代谢和物质代谢,有利于维持细胞内Ca2+的正常浓度。Objective To study the effect of Qinglongyi polysaceharide on Ca^2+ , Mg^2+-ATPase activity and [Ca^2+]i in erythrocyte of S180 mice. Methods S180 Mice were ip administrated with Qinglongyi polysaccharide for 7 d. The Ca^2+, Mg^2+-ATPase activity was observed by spectrophotometer with test kit and the [Ca^2+]i in erythrocyte of S180 mice was measured by laser scanning cofocal microscope (LSCM) with Fluo-3/AM. Results It showed that Qinglongyi polysaccharide increased Ca^2+, Mg^2+-ATPase activity (P〈0. 05), decreased the [Ca^2+]i in erythrocyte of S180 mice (P〈0.05) in a dose dependent manner. The effect in higher dosage was the best (P〈0.01). Conclusion Qinglongyi polysaccharide could increase the Ca^2+, Mg^2+-ATPase activity in erythrocyte of S180 mice, resolve the obstacle of ion transportion, stabilize the metabolism of energy and materials, and maintain the normal concentration of Ca^2+.
