摘 要:目的:研究喘息康对哮喘豚鼠模型肺泡灌洗液(BALF)中IFN-γ、IL-5及肺组织中基质金属蛋白酶-9(MMP-9)与其抑制剂(TIMP-1)的影响,探讨其治疗支气管哮喘的作用机制。方法:将40只豚鼠随机分组为正常对照组(A组)、哮喘模型组(B组)、喘息康中药组(C组)、地塞米松西药组(D组),每组10只。采用卵白蛋白(OVA)腹腔注射致敏和雾化激发的方法建立豚鼠支气管哮喘模型,然后以不同的方法对哮喘豚鼠模型进行干预。采用ELISA方法检测每只豚鼠BALF中IFN-γ、IL-5和肺组织中MMP-9、TIMP-1的变化。结果:哮喘模型组与正常对照组相比,前者BALF中IFN-γ水平显著降低(P<0.01),而IL-5则显著升高(P<0.01),肺组织中MMP-9、TIMP-1含量及比值显著升高(P<0.05~0.01);喘息康中药组和地塞米松西药组与模型组相比,BALF中IFN-γ水平高于哮喘模型组(P<0.01),而IL-5则显著降低(P<0.01),肺组织中MMP-9、TIMP-1含量及比值显著降低(P<0.05~0.01)。结论:喘息康可以通过促进哮喘豚鼠体内IFN-γ分泌和抑制IL-5表达,而降低支气管高反应性(BHR);并可通过降低肺组织MMP-9和TIMP-1含量,调节二者比值,抑制支气管哮喘豚鼠气道重塑。Objective:To investigate the effects of Chuanxikang decoction(a Chinese herbal medicine)on the amount of IFN-γ and IL-5 in bronchoalveolar lavage fluid(BALF) and expression of MMP-9 and TIMP-1 in lung tissue of asthmatic model in guinea pig to explore its mechanism in treatment of bronchial asthma.Methods:A total of 40 guinea pigs were randomly divided into 4 groups:normal control group(A group),asthmatic model group(B group),treatment with dexamethasone(C group) or with Chuanxikang decoction groups(D group),10 guinea pigs in each group.The guinea pig models of asthma were established through injecting ovalbumin(OVA) into the abdominal cavity and inhalation of atomization.The guinea pigs of asthma underwent with different therapy.Amount of IFN-γ and IL-5 in BALF and expression of MMP-9 and TIMP-1 in lung tissue were measured by immunotisssuchemical technology.Results:Compared with the normal control group(A group),the concentrations of IFN-γ in the BALF was decreased significantly(P〈0.O1)and IL-5 was increased significantly(P〈0.01) in the asthmatic model group(B group),the content and the relative value MMP-9 and TIMP-1 in lung tissues were also increased obviously(P〈0.05-0.01);IFN-γ in the BALF of the dexamethasone group(C group) and Chuanxikang decoction groups(D group)was increased significantly(P〈0.01) and IL-5 was decreased significantly(P〈0.01),the content and the relative value MMP-9 and TIMP-1 in lung tissues also were decreased obviously(P〈0.05-0.01) as compared with that in asthmatic model group(B group).Conclusion:Chuanxikang has therapeutic effect on bronchial asthma by increasing the level of IFN-γ and inhibiting the expression of IL-5 in the BALF;by decreasing the the content and the relative value of MMP-9 and TIMP-1 in lung tissue,which may lead to restrain airway remolding.
