摘  要:【目的】观察脑力智宝对学习记忆障碍模型小鼠的保护作用。【方法】(1)选用NIH小鼠32只随机分成假手术组,模型组,脑力智宝低、高剂量组(剂量分别为4、8.gkg-1·d-1);除假手术组外,其他各组小鼠均进行脑缺血再灌注损伤造模,中药组小鼠连续灌胃给药10 d后,采用跳台法观察小鼠的学习记忆能力,检测脑组织中丙二醛(MDA)与一氧化氮(NO)的含量、超氧化物歧化酶(SOD)与谷胱甘肽过氧化物酶(GSH-Px)的活力,高效液相色谱法检测大脑皮质中游离氨基酸的含量。(2)9月龄NIH小鼠40只随机分成正常对照组,拟衰老模型组,脑力智宝低、高剂量组(剂量分别为4、8.gkg-1.d-1);除正常对照组外,其他组均腹腔注射50 g/L D半-乳糖0.5 mL连续6周,复制拟衰老模型,同时灌胃给药连续6周后,采用跳台法测试小鼠的学习记忆功能,检测大脑皮质单胺类神经递质去甲肾上腺素(NE)、肾上腺素(E)及5-羟色胺(5-HT)的合成前体物质色氨酸(TP)的含量。【结果】(1)脑力智宝能显著提高脑缺血再灌注损伤小鼠的记忆能力(P<0.01),降低小鼠脑组织中MDA、NO、谷氨酸(G lu)的含量(P<0.05),提高SOD及GSH-Px的活力(P<0.05或P<0.01)。(2)脑力智宝能显著改善拟衰老模型小鼠的学习记忆能力(P<0.05或P<0.01),提高大脑皮质NE、E及TP的含量(P<0.05或P<0.01)。【结论】脑力智宝可以通过提高机体抗氧化酶活性,清除自由基,抑制兴奋性氨基酸与NO的神经毒性作用,调节大脑皮质单胺类神经递质含量等多种途径改善学习记忆功能。Objective To observe the protective effect of Naoli Zhibao (NZ) on mice with learning and memory impairment. Methods (1) Thirty-two NIH mice were randomized into pseudo-operation group, model group, and lowand high-dose NZ groups (at the dose of 4 and 8 g· kg^-1· d^-1 respectively). Except the pseudo-operation group, the mice in other groups received the operation of cerebral ischemia/reperfusion. After treatment for 10 days, the learning and memory abilities of the mice were observed by step-down test, and the contents of malondialdehydc (MDA) and nitric oxide (NO) as well as the activities of superoxidc dismustase (SOD) and glutathione peroxidase (GSH-Px) were detected. Meanwhile, the free amino acid content in mice cerebral cortex was measured with high performance liquid chromatography. (2) Forty NIH mice aged 9 months were randomized into normal control group, mimetic aging group, and low- and high-dose NZ groups (at the dose of 4 and 8 g· kg^-1· d^-1 respectively). The mice models of mimetic aging were induced by intraperitoneal injection of D-galactose 0.5mL for 6 weeks, and the mice in NZ groups were also given gastric gavage of NZ for 6 weeks. After treatment, the learning and memory abilities of the mice were observed by step-down test, and the contents of epinephrine (E), norepinephrine (NE) and tryptophan (TP) in mice cerebral cortex were measured. Results (1) NZ significantly improved the memory ability of mice with cerebral ischemia/reperfusion injury ( P 〈 0. 01 ) , reduced cerebral MDA, NO and glutamic acid (Glu) contents (P 〈 0. 05 ), and increased SOD and GSH-Px activities (P 〈0.05 or P 〈0.01 ). (2) NZ improved the learning a. d memory abilities of mimetic aging mice, and increased cerebral NE, E and TP contents (P 〈0. 05 or P 〈0. 01 ).
