摘  要:【目的】观察强肌健力饮对肾阳虚大鼠下丘脑—垂体—性腺轴作用的影响。【方法】将SD大鼠随机分为正常对照组,肾阳虚模型组,强肌健力饮低、中、高剂量组(剂量分别为7.8、15.6、23.4.gkg-1·d-1)、右归丸组(剂量为5.8.gkg-1·d-1),除正常对照组外,均采用氢化可的松(剂量为25 m.gkg-1·d-1)肌肉注射复制肾阳虚大鼠模型。观察动物的一般状态,计算下丘脑、睾丸指数并对睾丸做组织形态观察,采用放射免疫分析法检测血清睾酮(T)和雌二醇(E2)含量。【结果】强肌健力饮能显著降低下丘脑指数,升高肾阳虚模型大鼠血清T含量及降低E2/T比值(均P<0.01),而E2含量无明显变化(P>0.05);组织形态学观察发现,强肌健力饮各剂量组睾丸曲精小管内精细胞体积明显大于肾阳虚模型组,精原细胞核染色质和初级精母细胞核染色体比较显著。【结论】强肌健力饮防治中医肾阳虚证的作用机制可能与其能调控性腺轴的水平有关。Objective To observe the effect of Qiangji Jianli Yin (QjY) on hypothalamic-pituitary-gonad axis of rats with kidney-yang deficiency syndrome. Methods SD rats were randomized into normal group, model group and QJY groups (treated with QJY at the doses of 7.8, 15.6 and 23.4 g· kg^-1· d^-1 respectively), and Yougui Pill group (at the dose of 5.8 g· kg^-1· d^-1 ). Rat models of kidney-yang deficiency syndrome were induced by intramuscular injection of hydrocortisone 25 mg· kg^-1· d^-1. The outer appearance and activity of daily life of rats were observed, hypothalamus index and testis index were calculated, histological feature of testes was examined, and the serum levels of testosterone (T) and estrogen (E2 ) were measured by radioimmunoassay (RIA). Results In QJY groups, the hypothalamus index was decreased, serum T level were increased, and E2/T ratio was reduced (P 〈 0. 01 compared with the model group). The results of histological examination showed that the size of the spermatid in convoluted seminiferous tubule of QJY group was larger than that in the model group, and nuclear chromatin in spermatogonia (PL) and chromosome in primary spermatocytes were presented obviously in QJY group. Conclusion The preventive and therapeutic mechanism of QJY for rats with kidney-yang deficiency syndrome is probably related with the regulation of the hypothalamic-pituitary-gonad axis.
