长期储藏芍药花粉生活力测定方法研究

摘　　要：采用I2-KI染色法和TTC染色法测定储藏于室温、4℃、-20℃条件下的芍药花粉生活力,同时用培养基萌发法测定花粉萌发率,研究I2-KI染色法和TTC染色法测定结果与培养基萌发法测定结果的相关性。结果表明:芍药花粉生活力和萌发率随储藏时间的延长逐渐降低,且储藏温度越高,萌发率下降速度越快,室温下花粉储藏7d后萌发率即降为0,4℃储藏6个月后花粉萌发率降为0,-20℃储藏9个月后花粉萌发率降至1.96%。不同储藏温度下,I2-KI染色法、TTC染色法测定结果与培养基萌发法测定结果间呈极显著、显著相关;与TTC染色法相比,4℃储藏4个月内及-20℃储藏七个月内的花粉用I2-KI染色法测定的结果更为精确。The viability of Chinese peonies varieties pollen preserved in different temperature（room temperature,4 ℃,-20 ℃）and storage time were detected by I2-KI coloration and TTC coloration,and the germination rate of pollen were detected at the same time by germination in culture medium,to research the correlation of I2-KI,TTC and cultured in vitro.The results were as follow：the pollen viability and germination rate of Chinese peonies varieties reduced gradually as the storage time,and the descent of germination rate was quickest at room temperature,the germination rate of pollen preserved at room temperature for 7 days or 4 ℃ for 6 months reduced to zero,and the germination rate of pollen preserved at-20 ℃ for 9 months reduced to 1.96%.The correlation coefficient between I2-KI,TTC with germination in culture medium,respectively were extremely significant or significant correlate.Compared with TTC,the results of I2-KI in 4 ℃ preserved for 4 months and-20 ℃ preserved for 7 months were more accurate.
