摘 要:目的比较3种不同的固定和标记方法在流式细胞仪检测电离辐射与磷酸化H2AX(γ-H2AX)的剂量反应关系上的差异。方法用137Cs源分5个剂量点(0、1、2、4、8Gy)照射人乳腺癌细胞(MCF-7),分别用70%乙醇双标、70%乙醇单标和2%多聚甲醛单标MCF-7细胞,应用流式细胞仪检测电离辐射与γ-H2AX剂量效应关系。结果3种方法均发现电离辐射与γ-H2AX水平存在剂量反应关系,即γ-H2AX水平随着电离辐射剂量的增加而增加。拟合剂量反应曲线符合直线方程。乙醇固定双标法的决定系数最大,为0.87。结论应用流式细胞仪乙醇固定双标法检测MCF-7细胞电离辐射与γ-H2AX剂量效应关系,优于乙醇或多聚甲醛单标法。Objective To compare the differences among three different methods in evaluating the dose-effect relationship between ionizing radiation and H2AX by flow eytometry (gamma H2AX, γ-H2AX). Methods Human breast carcinoma cell MCF-7 was irradi- ated with 0, 1, 2, 4 or 8 Gy from ^137Cesium. Three methods, 70% ethanol-fixed with dual-staining, 70% ethanol-fixed with single- staining and 2% paraformaldehyde-fixed with single-staining were applied to evaluate the dose-effect relationship between ionizing radiation and γ-H2AX using flow cytometry. Results The dose-effect relationship between ionizing radiation and γ-H2AX could be determined by all the three methods, while the highest determination coefficient ( R2 ) was 0. 87, from 70% ethanol-fixed with dual-stai- ning method. Conclusions The results suggested that the method of 70% ethanol-fixed with dual-staining is better than other two methods in evaluation of dose-effect relationship between ionizing radiation and γ-H2AX using flow eytometry.
