摘  要:目的建立高效液相色谱(HPLC)法测定丹参提取物中水溶性成分丹参素、原儿茶醛和丹酚酸B含量的方法。方法高效液相色谱梯度洗脱法。色谱柱为Hypersil ODSC18色谱柱(250mm×4.6mm,5μm);流动相为1%冰醋酸溶液(A)和甲醇(B)梯度洗脱;检测波长为281nm;流速为1.0ml·min^-1;柱温25℃。结果丹参素、原儿茶醛和丹酚酸B线性范围分别在0.26~2.08μg,0.13~1.04μg,0.45~3.60μg内线性关系良好(r≥0.9996);加样回收率分别为97.76%,99.56%,99.68%,RSD〈3.4%。结论该方法操作简便、灵敏度高、重复性好,可用于丹参提取物质量控制。Objective To develop an HPLC method for determination of 3 water - soluble components - danshensu, protocatechuic aldehyde and salvianolic acid B in the extract from Salvia miltiorrhiza Bge. Methods The analysis was carried on with a column of Hypersil ODS C lS (250 mm× 4.6 mm ,5μ m) by HPLC at 25℃, with gradient elution of 1% (v/v) acetic acid (A) - methanol (B) as the mobile phase. The flow rate was 1.0 ml · min^-1 and the detection wavelength was 281 nm. Results The linear ranges of danshensu, protocatechuie aldehyde and salvianolic acid B were 0.26 - 2.08 μg, 0.13 - 1.04 μg and 0.45 - 3.60 p.g respectively, and all the compounds showed good linearity( r≥0. 999 6) in corresponding detected concentration range. The average recoveries were 97.76%, 99.56% , 99.68 % , and RSD was less than 3.4%. Conclusion The method is simple, sensitive, accurate, reproducible, and can be used to control the quality of extract from Salvia miltiorrhiza Bge..
