槐定碱对青霉素致痫大鼠中枢活动的影响

摘　　要：目的探讨槐定碱对青霉素致痫大鼠中枢活动的影响。方法大鼠50只,随机分为5组,每组10只,实验3组分别腹腔注射槐定碱2.94、5.88和11.76 mg/kg,阳性对照组和模型组分别腹腔注射苯巴比妥钠10mg/kg和生理盐水,观察槐定碱对大鼠癫痫模型大脑皮层脑电、脑组织形态学的影响及对其脑组织中MDA含量、SOD活性的影响。结果槐定碱2.94mg/kg可延长癫痫大鼠惊厥潜伏期,明显减轻大鼠痫样发作程度,明显减少大鼠皮层脑电图中的棘波;青霉素致痫大鼠后,光、电镜观察可见大鼠脑海马部位神经细胞肿胀,细胞器损伤等改变。使用槐定碱2.94mg/kg后神经元细胞肿胀明显减轻,细胞器等损伤能够得到不同程度的改善;槐定碱2.94mg/kg能降低癫痫大鼠脑中MDA含量,提高SOD活性;槐定碱5.88和11.76mg/kg未显示上述作用。结论槐定碱2.94mg/kg具有一定的抗癫痫作用,而槐定碱5.88和11.76mg/kg可能有一定的致痫作用。Objective To explore the effect of Sophoridine（SOP） on centre nerves system monement in the seizure rats induced by Penicillin. Methods Models of epilepsy was established by administration of Penicillin and epilepsy and related effects of SOP were observed. Results SOP of 2.94mg/kg dose significantly prolonged the latencies of epileptic seizure and significantly reduced seizure degree; SOP of 2.94mg/kg dose significantly changed seizure EEG; after epilepsy induced by penicillin,we could observe neuron tttmefaetion, organelle damnifieation and other changes. Sophoridine could significantly decrease neuron tumefaction and improve organelle damnification; SOP of 2.94rng/kg dose significantly decreased the MDA levels and significantly increased the SOD activity in the brain tissue of epileptic animals. Conclusion SOP of 2.94mg/kg dose can inhibit the epileptiform activity induced by penicillin, but SOP of 5.88mg/kg dose and SOP of 11.76mg/kg dose maybe have opposite effect.
