摘 要:目的建立犬血浆中甲硝唑浓度反相高效液相色谱(RP-HPLC)测定方法。方法采用反相高效液相色谱法C18柱,以0.05mol/L的KH2PO4(用1mol/L的磷酸调节pH3.5)∶乙腈∶甲醇(130∶35∶35,V/V/V)作为流动相,流速0.6ml/min,检测波长317nm,柱温25℃,内标为替硝唑。结果RP-HPLC测定甲硝唑血药浓度的线性范围是0.2-20.0μg/ml(r2=0.9986),定量下限为0.2μg/ml,准确度在85%-115%范围内,其日内、日间精密度≤15%。结论本法可用于甲硝唑犬体内药动学研究的需要,并成功地用于口服甲硝唑片剂后犬体内的药动学研究。Objective To establish a RP-HPLC methood for determining the plasma concentration of metronidazole in dog. Methods A RP-HPLC method was developed. A column of C18 was used, with 0.05 mol/L KH2PO4 : acetonitrile: methnol( 130:35:35, V/V/V) as a mobile phase. The flow rate was 0.6 ml/min. The detective wavelength was set at 317 nm,and the column temperature was 25 ℃. Tinidazole was used as internal standard(IS). Results The calibration curve was linear over the concentration range of 0.2 - 20.0 μg/ml. Lower limit of quantification(LLOQ) was 0.2μg/ml, and the accuracy of metronidazole was 85 % - 115 %. The precision RSD was less than 15 %. Conclusion The RP-HPLC is validated to guarantee a reliable determination of metronidazole concentration in dog plasma. The method can be applied to investigate the pharmacokinetics profile of metronidazole in dogs after oral administration.
