摘 要:初步研究了分离水稻小孢子原生质体的条件,首次从具小萌发孔的水稻小孢子中分离出原生质体.小孢子原生质体的分离途径是:小孢子在混合酶液作用下,质膜先脱离小孢子壁,然后原生质体缓慢地从萌发孔中挤出.混合酶液中的渗透压高低和分离时的温度条件对小孢子原生质体的分离有很大影响,在24℃温度和0.6mol/L甘露醇的渗透压下分离效果最佳,此时原生质体的分离率达到9.8%~11.2%.It might be the first success to isolate the protoplast from microspore in rice. The conditions for isolating microspore protoplast were studied. The process for microspore protoplast isolation was as follows: first, plasm membrance seperated from the microspore wall, then the protoplast inside microspore was partially extruded through the germinal hole after the full action of enzyme solution, and finally being completely detached from the exine of microspores, a free & spherical protoplast was formed. Microspore protoplast isolation was greatly influenced by the mannitol concentrations in enzyme solution and the degree of incubation temperature, the appropriate mannitol concentration and temperature seemed to be 0.6 mol/L mannitol in enzyme solution and incubated at 24 ℃ temperature. In this experiment, the frequency of protoplast isolation reached as high as 9.8%~11.2%.
