三七总苷对自身免疫性睾丸炎大鼠中TNF-α表达与定位的影响

摘　　要：目的:研究三七总苷对自身免疫性睾丸炎睾丸组织中TNF-α表达和血清中含量的的影响,探讨三七总苷通过调控TNF-α分泌来拮抗睾丸炎进展的作用及作用机制。方法:参照王涛等人的方法建立经典的自身免疫性睾丸炎模型,随机分为模型组和三七总苷低、中、高剂量预防组各10只,免疫80天后取材,ELISA法测血清TNF-α含量,免疫组织化学方法定位TNF-α在睾丸组织中的表达。结果:模型组血清TNF-α含量显著高于正常对照组(p<0.05);模型组间质炎症细胞浆、生精小管基底膜、多核巨大精细胞TNF-α呈强阳性表达,生精上皮表达较弱,三七总苷中、高剂量预防组可明显降低血清中TNF-α的含量及在睾丸组织中的表达强度。结论:三七总苷中、高剂量预防组可能通过调控过量TNF-α的表达,进一步拮抗自身免疫性睾丸炎的进展。Objective： To observe the effects on panax notoginseng saponins （PNS） on expression and levels of TNF - α in EAO of rats and to explore its mechanisms on antagonizing orchitic progression. Methods： Duplicated EAO model, Sprague - Dawley rats were divided randomly into model group and PNS lower - , medium and high - dosage preventing groups （ ten rats, individually）. After 80 days,the levels of serum TNF - α were measured by ELISA, localizations of TNF - α in testicular tissue were by immunochemistry （IHC）. Results： In model group testicular TNF - α expressions were positive significantly in the cytoplasms of the interstitial cells, basal laminas and polynuclear sperm cells. In model group the levels of TNF - α were significantly higher than those in the normal group（ p 〈 0. 05）. PNS medium - and high - dosage preventing groups decreased obviously on levels and expression of TNF -α. Conclusion： PNS medium - and high - dosage preventing groups could postpone the progression of EAO through down - regulation of TNF - α expression and secretion.
