摘 要:从大豆胚轴提取大豆异黄酮和皂甙,研究大豆异黄酮和皂甙抑制结肠癌细胞的增殖和促进其凋亡的作用。采用MTT比色法观察大豆异黄酮和皂甙抑制结肠癌细胞增殖,采用流式细胞仪检测大豆异黄酮和皂甙对HT-29细胞周期分布的影响,免疫细胞化学法检测凋亡相关蛋白bax、bc1-2和p53的表达。结果表明:大豆异黄酮和皂甙可时间和浓度依赖性地抑制结肠癌细胞增殖;诱导细胞凋亡和改变细胞周期分布,多数细胞阻滞于G2/M期;与对照组比较,细胞凋亡率显著增加;用药前后凋亡相关基因bax蛋白表达显著增加,bc1-2表达显著降低。提示,大豆异黄酮和皂甙可通过改变细胞周期分布和诱导结肠癌细胞凋亡,发挥抗结肠癌作用。Soybean isoflavones and saponins have been observed to limit many kind of tumor cell proliferation by inducing apoptosis. Soybean isoflavones and saponins were extracted from soybean hypocotyl, then investigated for the inhibition on proliferation and apoptosis of human colon carcinoma HT-29 cell. MTT assay was used to examine the proliferation of HT- 29, flow cytometry was used for analysis of cell cycle distribution, and immunohistochemical method was used for the determination of bax,bcl-2 and p53 expression. The results showed that soybean isoflavones and saponins inhibited the proliferation of colon carcinoma cell in a time- and concentration-dependent manner,induced apoptosis and lead to G2/M arrest. The apoptotic ratio of treatment group was significantly lower than that of control group, the expression of bax increased and bcl- 2 decreased significantly. It is suggested that soybean isoflavones and saponins could exert an anticolon cancer effect via cell cycle arrest and induction of apoptosis.
