摘 要:以超临界CO2(SCCO2)为分散介质,在聚偏氟乙烯(PVDF)微孔膜上进行马来酸酐和苯乙烯的接枝共聚,合成出超高相对分子质量(Mw=8.4×106)的苯乙烯/马来酸酐交替共聚物(SMA)基微孔PVDF膜。吸附实验表明,接枝率增大,接枝膜对蛋白吸附减少,抗污染性增强;细胞迁移实验中,接枝膜上细胞迁移范围及数量远高于原膜,生物相容性好;细胞粘附实验说明,接枝膜有助于细胞的粘附生长;细胞活性实验显示,细胞在接枝膜表面经3d培养后的乳酸脱氢酶活力低于4.5U/mL,说明接枝膜无细胞毒性;接枝膜动物急性实验中,未出现小白鼠死亡、瘫痪和呼吸困难等中毒现象。因此,接枝膜作为血液透析膜材料具有潜在应用前景。Surface modification of poly ( vinylidene fluoride ) (PVDF) microporous membrane by graft copolymerization with maleic anhydride (Man) and styrene (St) was carried out in supercritical carbon dioxide(SCCO2) system. The alternative copolymer segments were grafted onto the membrane surface and within membrane pores to obtain the SMA-based PVDF membrane(Mw = 8.4×10^6). Modification by SMA grafting made the concentration of the protein adsorption on the modified membrane decreased significantly, relating with excellent hydrophilicity of the SMA-based membrane. However, SMA-based membrane behaved preferable cell proliferation for cell growth after a given time of incubation, no induced toxicity of cells attached to the modified membranes was found, and the amount and ambit of the migratory cell from rabbit cornea to SMA grafted membrane behaved preferable than the original membrane. This also showed excellent biocompatibility of the SMA-based membrane. After three days cultivation,the lactic acid dehydrogenase (LDH) released from the attached cells has activity lower than 4. 5 U/mL and the Mus musculus albus injected with SMA grafted membrane soak has good growth condition after seven days observation in the cytotoxicity tests. This revealed that the SMA grafted membrane has no cytotoxicity and destructiveness to the attached cells. All these can create new opportunities for SMA-based PVDF membrane for use as a hemodialysis material.
