摘 要:目的:控讨幼年大鼠肾小管间质损伤早期,跨膜信号传导通路中JAK2/STAT3信号分子表达趋势及其与转化生长因子-1β(TGF-1β)表达的相关性。方法:以单侧输尿管梗阻(UUO模型)幼年大鼠为实验模型;采用免疫组织化学方法检测,行UUO术后3,7,14,28 d大鼠肾组织JAK2/STST3及TGF-1β蛋白的表达,并从时相上评价JAK2/STAT3与TGF-1β表达的相关性。结果:UUO组3 d各指标表达强度即增加,随着时间的延续,三者呈同步上调趋势。各时间点与对照组比较有统计学差异(P<0.01);UUO组内各时间点间比较有统计学意义(P<0.01),各指标两两之间的相关系数r值均大于0,且呈高度正相关关系。结论:UUO大鼠模型肾小管间质损伤早期,JAK2/STST3及TGF-1β间可能存在一条促肾小管质损伤的信号途径。Objective:To investigate the trend of expression and relatinvity of JAK2/STAT3 signal protein and TGF-β1 on early stage tubulointerstitial lesions in young rats of unilateral ureteral obstruction(UUO). Methods :Male young wistar rats with unilateral ureteral obstruction were used as experimental models. The JAK2/STAT3 and TGF-β1 protein expression in renal tissue were detected by immunohistochemical ways on days 3,7,14 and 28 after UUO and the correlation among JAK2/STAT3and TGF-β1 protein expression in time phase were evaluated. Resuhs:Immunohistochemical staining indicated that the level of expression of the three proteins were increased at day 3 in UUO group compared with control group. And they were up-regulated simultaneously in tubular epithelial cells. The significant difference were observed between control groups and UUO groups in different time points (P〈0.01). Positive correlations were observed among the three factors (P〈0.01). Conclusion:There might be a biologic signalpathway among JAK2/STAT3and TGF-β1 in the early phase of. tubulointerstitial lesions induced by unilateral ureteral obstruction in young rats ,also,it might be one of mechanism that enhanced tubulointerstitial nephropathy.
