摘 要:目的:探讨多孔状生物活性玻璃陶瓷(bioactive glass ceramics,BGC)与体外培养扩增的兔骨膜源性成骨细胞(periosteal-derived osteoblasts,POBs)的生物相容性,为骨组织工程载体材料的选择提供依据。方法:体外培养并扩增兔骨膜源性成骨细胞,运用MTT法和酶学测定法分别检测BGC提取液对细胞增殖和碱性磷酸酶(ALP)活性的影响,运用倒置相差显微镜、扫描电镜观察与BGC复合培养的细胞在材料内的生长情况。结果:BGC提取液对培养早期细胞的增殖有一定的抑制作用,对培养晚期细胞的增殖无明显影响;在细胞培养过程中能上调POBs分泌ALP的能力;细胞在BGC材料孔隙表面生长良好,并能增殖、分泌细胞外基质。结论:在体外多孔状BGC具有良好的细胞相容性,为BGC作为骨组织工程的细胞载体提供了实验依据。Objective: The purpose of this study was to investigate the biocompatibility of the bioactive glass ceramics (BGC) combined with cultured periosteal-derived osteoblasts(POBs) in vitro, and to select a favorable scaffold for bone tissue engineering. Methods: POBs harvested from a new born rabbit were cultured with the supernate of BGC, at different time points, cell proliferation ability and alkaline phosphatase(ALP) activity were examined by MTT method and enzyme assay. The morphological characters of POBs seeded into the porous BGC were observed by scanning electronic microscope (SEM). Results: The supernate of BGC had a little negative effect on the proliferation of POBs. BGC could stimulate the POBs to secrete more ALPs. Most of the seeded cells grew well on the surface and could produce some extracellular matrix (ECM). Conclusion: BGC had an excellent biocompatibility and osteoconduction with cultured POBs in vitro, and could be used as scaffolds in bone tissue engineering.
