现代罗布人mtDNA 9bp序列缺失频率与DYS287位点多态性研究

摘　　要：目的:研究现代罗布人线粒体DNA 9bp序列缺失频率与Y染色体DYS287位点多态性。方法:分别采用PCR扩增直接测序法和PCR结合琼脂糖凝胶电泳检测法对不同位点的多态性进行分析。结果:在48名无关现代罗布人群个体中,线粒体DNA9bp序列缺失频率为8.3%,在34名无关现代罗布人群男性个体中DYS287全部显示为YAP-。结论:获得现代罗布人线粒体DNA 9bp序列缺失频率与Y染色体DYS287位点多态性数据,为该群体遗传关系的分析、法医学鉴定及该群体的起源提供了一定的遗传背景资料。Objective： To investigate the frequency of 9bp deletion of mtDNA and the genetic polymorphism of the Y- chromosome specific Alu insert sequence DYS287 among Modem Lopnur People. Method： Respectively adopt PCR directly sequensing method and PCR combine with agarose gel electro - phoresis to detect Polymorphism of the different sites in the product. Result： Among 48 unrelated people, The frequencies of 9bp deletion in mtDNA in Mordem Lopnur People is 8.3 %, Among 34 unrelated males, YAP element was absent. Conclusion： The genetic data of 9bp deletion of mtDNA and DYS287 loci in Mordem Lopnur People have been obtained, and they might be useful in studies of population origin, relationship between populations, and forensic applications.
