摘 要:目的:一株饵料微藻与益生菌混合固定化培养条件的优化。方法:在单因素实验基础上,采用U6*(64)均匀设计表,对海藻酸钠浓度、氯化钙浓度、藻的接种量和菌的接种量进行优化实验,并采用DPS软件对实验结果进行分析,以获得适宜的混合固定化条件;并按照此优化条件对混合固定化和单藻、单菌固定化胶球中微藻和益生菌的生长速率进行比较。结果:①获得适宜的混合固定化条件为:海藻酸钠浓度4%,氯化钙浓度1.2%,藻接种量为21.9889×107个细胞,菌接种量为58.7676×109个细胞;②混合固定化胶球中藻细胞的生长速率比单藻固定化胶球中的藻的生长速率提高了18.8%;菌的平均生长速率比单菌固定化胶球中的菌的平均生长速率提高了92.6%。结论:该实验首次对饵料微藻和海洋益生菌的混合固定化培养条件进行优化。发现藻菌混合固定化胶球中藻和菌的生长速率比单独固定化均有较明显提高。实验结果说明,在混合固定化的微环境体系中,特定的藻菌株具有相互促进生长的作用。实验结果为藻菌混合固定化技术的应用和发展提供了必要的实验数据和理论基础。Objective: The optimization of the co - immobilization conditions of a bait microalgae and probiofics. Method: In the base of single factor experiments,uniform design was used to optimize the the concentration of SA, CaCl2, the number of algae inoculation and prohioties. The immobilization conditions which fit both algae and prohiotios were expected.And the experimental results were analyzed by Data- processing System (DPS).Bait microalgae and probioties'grewth speed in the co-immobilized beads were compared with single immobilized beads. Result: Firstly, the optimum co- immobilization condifiom: The concentration of SA was 4%, the concentration of CaCl2 was 1.2%, the number of algae inoculation was 21.9889 ×10^7 cells,the number of probiofies inculation was 58.7676 ×10^9 cells.Secondly,the algae cells' growth speed in co - immobilized beads was 18.8% higher than in single algae immobilized beads,and the probiotics cells' growth speed in co- immobilized beads was increased 92.6% compared with which in single probiotics immobilized beads. Conchusion:The experiment was used to optimize the conditions which mostly affect bait microalgae and marine probiotics in the co-immobilization incubation for the first time. Through which algae and probioties' growth speed increased evidently were found. And that given algae and bacteria could accelerate development each other in co - immo- bilized micro- environment systerm was showed in this experiment.Essential experiment datum and theoretical base to the application and development of algae- bacterial co- immobilization were offered in the results of this experiment.
