摘 要:为尝试构建基于质谱分析的家蚕蛋白质组数据库,采用一维电泳-液相色谱-质谱(1DE-LC-MS)技术对家蚕蚁蚕的蛋白质组进行了分析与鉴定。共检索到511个相匹配的候选蛋白质和92个多肽离子片段,鉴定获得了171个无冗余蛋白质组分,分别属于121种蛋白质或蛋白质亚基,其中有36种蛋白质或亚基尚未在家蚕及昆虫的基因与蛋白质数据库中报道;在171个被鉴定的蛋白质组分中,与细胞骨架和蛋白质代谢相关的组分多达64种。结果还表明,1DE-LC-MS技术也适用于从一维凝胶电泳的差异性条带及蛋白质混合样品中分离和鉴定特异蛋白质组分。In order to construct the proteome database based on mass spectrometry in the silkworm, Bombyx mori, the full proteins of newly-hatched silkworm were analyzed and identified by method of one-dimensional electrophoresis-liquid chromatography-mass spectrometry (1 DE-LC-MS). The results showed that 511 matchable protein candidates and 92 peptide ion-fragments were obtained. By eliminating the redundant protein candidates, a total of 171 protein compositions were identified and defined as 121 kinds of proteins or protein subunits. Among them, 36 kinds of proteins have not been deposited in the DNA or protein database of silkworm as well as other insects. The feature of the protein compositions showed that more than 64 components among 171 identified proteins were relevant to cell structure and protein metabolism. In addition, the results also showed that the 1 DE-LC-MS technique is also suitable to separate and identify target proteins from the differential bands of one-dimensional gel electrophoresis and the composite protein samples.
