摘 要:目的探讨活血通脉灵对实验性心肌缺血大鼠心肌损伤的抗黏附保护作用机制。方法将肢体II导联心电图正常的Wistar雄性大鼠50只随机分为5组,每组10只(健康对照组、模型对照组、消心痛对照组及活血通脉灵大剂量治疗组和小剂量治疗组)。健康对照组以普通饲料喂养,余4组以高脂饲料喂养并均建立心肌缺血模型,并且均灌胃干预。后取心尖部心肌组织,进行苏木素—伊红(HE)染色,并采用ABC染色检测P-选择素(P-Selectin)和L-选择素(L-Selectin)的表达量。结果与健康对照组比较,其余4组大鼠P-Selectin和L-Selectin表达均明显升高,差异有统计学意义(P<0.05或P<0.01),与模型对照组比较,各治疗组都有不同程度的降低,其中大剂量组差异最大,消心痛对照组和小剂量组差异较小,但均有统计学意义(P<0.01)。消心痛对照组和小剂量治疗组间差异无统计学意义(P>0.05)。结论活血通脉灵对大鼠心肌缺血引起的心肌损伤具有保护作用,减少缺血心肌P-Selectin、L-Selectin的表达,有一定抗黏附作用。Objective To study the protective effect and mechanism of Huoxuetongmailing (HXTML) on experimental myocardial ischemic rats. Methods 50 healthy male Wister rats were randomly divided into five groups ( 10 rats in each group), namely blank group, model group, Cedocard group, high-dose HXTML and low-dose HXTML group, of which limb II electrocardiography (ECG) were recorded before the experiment. The rats were fed with hyperlipidemic forage and ISO was subcutaneously injected into rats except blank group to cause myocardial ischemia. After treatment with the chugs noted above, HE staining was employed to analyze the pathological damage and ABC staining to detect the expression of P-Selectin and L-Selectin in myocardium of heart apex. Results P-Selectin and L- Selectin expression were significantly higher in myocardium of heart apex in the four groups when compared with the blank group( P 〈0.05 or P 〈 0.01) .They were lower in treatment groups than model group( P 〈 0.01).While there were no significant difference existed between low-dose group and Cedocard group ( P 〉 0.05). Conclusion The HXTML could alleviate the myocardial damage and protect myocardial tissue of rats. Furthermore, it had some particular effects on P-Selectin and L-Selectin to lower blood viscosity.
