摘 要:目的研究细胞外铁离子(Fe3+)浓度变化对人成骨细胞(hFOB 1.19)内Ca2+转运的影响。方法将成骨细胞34℃培养3~4 d(细胞密度90%)并用胰蛋白酶消化后分种于12孔培养板上。空白组加入双蒸水;实验组加入不同浓度的去铁氨(DFO)和枸橼酸铁铵(FAC)。分别干预后立即用激光共聚焦扫描显微镜(CLSM)检测。结果CLSM显示,随着hFOB 1.19外Fe3+浓度的减少(DFO终浓度为200~300μmol/L),Ca2+向细胞内的转运量增加;而随着hFOB 1.19外Fe3+浓度的增加(FAC终浓度为50~100μmol/L),Ca2+向细胞内的转运呈降低趋势。结论hFOB 1.19外Fe3+浓度的减少可以增加细胞内的Ca2+,而胞外Fe3+浓度增加则减少细胞内的Ca2+。Objective To study the influence of iron ion on calcium transportation of human osteoblast (hFOB1.19 cells). Methods The hFOB 1.19 cells were cultured at 34℃ for 3-4 days, digested with trypsin, and then put on a 12-hole cultivate board. D2 H2O was added in control group, and different concentrations of DFO or FAC were used in experiment group. Confocal laser scanning microscope (CLSM) examination was performed immediately. Results CLSM examination showed that the diversion of Ca2+ into cells was increased as the decrease of Fe3+ concentration out of the hFOB 1.19 cells(final concentration of DFO,200~300μmol/L) ,which was decreased as the increase of Fe3+ concentration out of them(final concentration of FAC, 50~100μmol/L). Conclusion The Ca2+ transportation towards cells was increased with the decrease of Fe3+ concentration out of hFOB 1.19 cells,which was decreaced as the Fe3+ concentration increased.
