摘 要:采用蛭石栽培,对开花后10 d的菜用大豆[Glycine max(L.)Merr.]用100 mmol.L-1NaCl进行10、15、20 d胁迫处理,利用双向电泳技术对种子蛋白质进行分离,并对处理与对照图谱中β-伴大豆球蛋白各亚基和大豆球蛋白各酸性肽的表达进行比较分析。结果表明:胁迫10 d时α、α′亚基及A1a、A4酸性肽的丰度均显著下调;胁迫15 d时α、β亚基及A4酸性肽的丰度均显著上调,A1a、A3酸性肽则显著下调;胁迫20 d时A1a的丰度显著上调,α、β、A2、A3、A4均显著下调。尽管有些亚基或多肽的丰度在个别时期显著升高,但大豆球蛋白的整体表达量在NaCl胁迫期间均显著下降。结果表明,NaCl对菜用大豆球蛋白的积累在各个胁迫时期均有显著的抑制作用,随着胁迫时间的延长受抑制的亚基及多肽增加。Under vermiculite culture conditions, vegetable soybean Glycine max (L.) Merr. was treated with 100 mmol.L^-1 NaCI at the 10th day after flowering. The proteins from the seeds at the 10th,15th and 20th days under NaCl stress were seperaped with two-dimensional gel electrophoresis (2-DE). The expression differences were analyzed in β-conglycinin subunits and glycinin acidic polypeptides between treatment and the control. The results showed that the abundances of α,α',A1α and A4 were all down-regulated significantly at the 10th day of salinization. At the 15th day of NaCl stress, the abundances of α,β and A4 increased remarkably,while those of A1α and A3 decreased significantly. At the 20th day of NaCl treatment, the abundance of A1, was up-regulated significantly, while the abundances of α, β, A2, A3 and A4 were downregulated remarkably. Though the abundances of either subunits or acidic polypeptides were increased significantly at a certain period,the overall expression levels of globulins in the seeds were decreased significantly during NaCl stress period. These results indicated that the accumulation of seed globulins of vegetable soybean was inhibited remarkably during NaCl stress period,and the numbers of both subunits and acidic polypeptides with inhibited expressions were increased when the stress time was prolonged.
