摘  要:目的:探讨加减抵挡汤治疗肾小球硬化的机制。方法:采用单侧肾切除重复阿霉素注射造成肾小球硬化大鼠模型,以加减抵挡汤治疗并用西药科素亚为阳性对照,观察模型组和各治疗组的肾功能及24h尿蛋白定量,采用逆转录多聚酶链反应(RT-PCR)测定各组大鼠肾组织TIMP-1及其PAI-1的mRNA的表达。结果:加减抵挡汤各组均能降低肾小球硬化大鼠增高的24h尿蛋白,改善肾功能,且加减抵挡汤能下调大鼠肾组织TIMP-1和PAI-1mRNA的表达。结论:加减抵挡汤通过下调肾小球硬化大鼠肾组织TIMP-1和PAI-1 mRNA的表达,延缓肾功能衰竭。Objective: To discuss the influence of Didang Decoction on glomerulosclerosis. Methods: The glomemlosclerosis model was established by uninephrectom and injection of adriamyci, and was treated by modified Didang Decoction and taken western medicine Losartan as the comparison group. To observe 24-hour urinary protein and kidney function of model group and each treatment group. Reverse transcriptase Polymerase Chain Reaction(RT-PCR) was used to determine the expression of TIMP-1 and PAI-1 in each group of rats kidney tissue. Results: Didang Decoction could decline elevated 24-hour urinary protein and improve kidney function, and decline the expression of TIMP-1 and PAI-lmRNA in glomerulosclerosis rats. Conclusion: Didang decoction could delay renal failure, it may be related to the down-regulated expression of TIMP-1 and PAI-1mRNA.
