摘  要:目的:卵巢激素对子宫内膜离子转运的影响及其可能机制。方法:采用短路电流(Isc)技术,体外测量动情期大鼠(正常对照组)及去卵巢大鼠(模型组)子宫内膜电流及电阻的变化。结果:模型组的基础Isc及跨膜电阻(R)分别为25.56±7.58μA/cm2和64.57±5.44?/cm2,均高于对照组(8.91±3.18μA/cm2;48.48±4.66?/cm2,P<0.05)。使用上皮Na+通道阻断剂阿米洛利对基础电流无明显影响;cAMP依赖的Cl-通道(CFTR)激动剂弗斯可林(10μmol/L)使Isc略有增加;CFTR阻断剂格列本脲使对照组的基础电流略有降低(P>0.05),而模型组则大幅度降低(P<0.05)。Ca2+激活的Cl-通道(CACC)阻断剂DIDS可使各组的Isc均降低,组内用药前后比较,P均<0.05;而Na+-K+-2Cl-共转运体的阻断剂布美他尼(100mmol/L)对二组Isc无明显影响。结论:去卵巢大鼠的子宫内膜的CFTR和CACC对阻断剂的敏感性均强于对照组,说明上皮离子的转运与卵巢分泌的雌、孕激素有关。Objective: To investigate the property of the endometrium ion transport in the normal estruation and ovariectomize rat. Methods: The basic electrophysiological property of the endometrium was carried out by mean of the short circuit current (Isc) recording with the model rats. Results: The baseline Isc and the transepithelial resistance (R) were 8.91±3.18 μAcm^2 (n=13) and 48.48±4.66 Ω/cm^2 (n=16) in the control, 25.56±7.58 μA/cm^2 (P〈0.05, n=12) and 64.57 ±5.44 Ω/cm^2 (P〈0.05, n=15) in the ovariectomized group. Apical application of Na^+ channel blocker, amiloride (10 μmol/L), did not significantly affect the Isc in the two groups. Application of the endometrial epithelium with the CPTR Cl^- channel agonist, forskolin (10 μmol/L), did not significantly increase the Isc in the two groups. Moreover, the Isc in the model group was more sensitive to the CFTR Cl^- channel blocker (glibenclamide, 1 mmol/L, apical side) than that in the control. Apical application CACC Cl^- channel blocker, DIDS (4,4'- diisothiocyanostilbene-2, 2'-disulfonic, 100 μmol/L), the Isc in the model group was much more sensitive than that in the control. However, basolateral application of bumetanide (100 μmol/L, basolateral), a blocker of Na^+-K^+-2Cl^- cotransport, didn't significantly reduce the Isc in the two groups. Conclusion: The Isc baseline and membrane resistance in the model group is much higher than that in the control, and the endometrial epithelium Cl^- secretion may be mediated by predominantly calcium-dependent Cl^- channels (CACC) and activating adenylate cyclase and apical cAMP-dependent Cl^- channels (CFTR) in the model group with minor contributions in the control. Furthermore, the anion transport in endometrial epithelium is related to estrogen and progestin which secreted by ovary.
