摘  要:采用PCR-RFLP技术对中国荷斯坦牛生长激素受体(GHR)基因第8外显子进行单核苷酸多态性分析。结果表明,GHR基因经TasⅠ酶切后产生2种等位基因和3种基因型。等位基因A和T的频率分别为0.6339和0.3661。基因型AA、AT和TT的频率分别为0.4558、0.350和0.191。测序分析表明TT型和AA型相比在GHR基因第4962位碱基处发生T→A突变,该突变导致氨基酸由苯丙氨酸(TTT)变为酪氨酸(TAT)。χ2适合性检验表明,GHR基因的TasⅠ酶切位点处于Hardy-Weinberg不平衡状态(P<0.05)。最小二乘分析表明:基因型TT所对应的乳脂率最小二乘均值显著高于基因型AA所对应的最小二乘均值(P<0.05)。Single nucleotide polymorphisms (SNPs) of exon 8 of growth hormone receptor (GHR) gene were detected in Chinese Holstein cows by PCR-RFLP. The results showed that the exon 8 of GHR gene digested by Tas Ⅰ could be divided into two kinds of alleles and three kinds of genotypes. The frequencies of allele A and T were 0.6339 and 0.3661 respectively. The frequencies of genotype AA, AT and TT were 0.458, 0.350 and 0.191, respectively. Sequencing revealed that one single nucleotide mutation T→A at 4 962 bp in genotype TT compared with genotype AA resulted in an amino acid change ofphenylalanine (TTT)→tyrosine(TAT). The result of Chi Square test indicated that the frequencies of pattern types of GHR gene digested by Tas Ⅰ were not fit with Hardy-Weinberg equilibrium in this population (P 〈 0.05). The least squares means of fat percentage in genotype TT were higher than that in genotype AA (P〈 0.05).
