摘  要:类钙调磷酸酶B互作蛋白激酶(calcineurin B-like protein interacting protein kinase,CIPK)在植物非生物胁迫信号转导中起重要作用。应用TR-PCR的方法在玉米(Zeamays)中克隆了一个cDNA全长为1395bp的蛋白激酶基因ZmCIPK1(GenBank登录号为EF158034)。其编码的蛋白含有465个氨基酸,分子量为51.98kD,等电点为6.92。推断的ZmCIPK1催化结构域中包含蛋白激酶催化结构域保守的11个亚结构域,蛋白C端具有植物CIPK蛋白家族特有的包含24个氨基酸的NAF结构域。半定量RT-PCR结果表明,在甘露醇、氯化钠、ABA和4℃低温诱导时,ZmCIPK1在玉米幼苗叶片中表达量明显上调,在24h内保持较高的转录水平。ZmCIPK1是一个新的玉米蛋白激酶基因,响应多种非生物胁迫信号。Calcineurin B-like protein Interacting (CIPK) protein kinase plays an important role in stress signal transduction in plant. One full-length eDNA encoding a CIPKs homologue was isolated from maize (Zea mays) by RT-PCR and named as ZmCIPK1 (GenBank accession No.EF158034). The predicted ZmCIPK1 protein had 465 amino acids with an estimated molecular mass of 51.98 kD and an isoelectric point of 6.92. The putative kinase catalytic domain of ZmCIPKI contained 11 subdomains that were typical in protein kinase. The conserved 24-amino acid motif within C-terminal non-kinase region of the CIPKs was also found in ZmCIPKI. RT-PCR analysis indicated that ZmCIPK1 expression was up-regulated obviously by mannitol, salt, ABA and low-temperature (4 ℃) in young leaves of maize and kept high level during 24 h. These results demonstrate that ZmCIPKI is a novel CIPK gene and responds to multiple abiotic signals in maize.
