大白菜-结球甘蓝4号单体异附加系的鉴定

摘　　要：大白菜-结球甘蓝异附加系是利用结球甘蓝(Brassica oleraceaL.var. capitata)的优良基因对大白菜(Brassica compestris L.ssp.pekinensis)进行品种改良的中间材料。研究以大白菜-结球甘蓝异源三倍体(AAC,2n=29)与二倍体大白菜(AA,2n=20)回交一代的自交后代为材料,经过细胞学鉴定,从中筛选出大白菜-结球甘蓝4号单体异附加系CO-4-1。终变期联会以10个二价体和1个单价体(10Ⅱ+Ⅰ)为主,约占85%;中期Ⅰ,绝大多数细胞中有染色体游离于赤道板外;后期Ⅰ多以11-10方式分布;后期Ⅱ多以11-10-11-10形式分离。叶背有轻微腊粉,叶面光滑无皱褶以及抽薹晚等田间性状与结球甘蓝相似。减数分裂和田间性状均可以说明额外染色体为甘蓝染色体。该异附加系的获得为研究芸薹属A、C基因组的亲缘关系,以及向大白菜中导入结球甘蓝的优良基因,扩大种质遗传背景具有重要意义。Chinese cabbage - head cabbage alien addition lines are middle breeding materials for improving Chinese cabbage （Brassica compestris L. ssp. pekinensis） using head cabbage （Brassica oleracea L.var. capitata） genes. In this study, a monosomic addition line named CO-4-1 （Chinese cabbage with no. 4 chromosome fi＇om cabbage） derived from the progenies of the Chinese cabbage-cabbage allotriploid （AAC, 2n=29） x Chinese cabbage diploid （AA,2n=20） was obtained by cytology identification. Chromosome configurations were mainly 10 Ⅱ ＋ Ⅰ with the percentage of 85% at diakinesis. The chromosome out of equator plank was observed in most of cells at metaphase Ⅰ . And most of the chromosome distribution was 10-11 at anaphase Ⅰ and 11-10-11-10 at anaphase Ⅱ. The field characters seemed like those of head cabbage with waxy leaf surface, leaf smoothness and late bolting time. The pollen-mother-cell＇ s meiotic chromosome behavior and the field characters indicated that the additional monosome belonged to head cabbage. It may be useful to explore the relationship between A and C genomes, transmit genes to Chinese cabbage, broaden genetic background of Chinese cabbage and increase the germplasm genetic diversity using these alien addition lines.
