摘  要:【目的】检测急性四氯化碳(CCl4)中毒大鼠肝细胞中整合素α1(Integrinα1)的表达,探讨五味子在治疗急性肝损伤时对于Integrinα1表达的影响及意义。【方法】将Wistar大鼠随机分为对照组、中毒组和治疗组。中毒组和治疗组大鼠经腹腔一次性注射20%的CCl4(按0.2 mL/100 g剂量)致毒。治疗组以五味子提取液每天(0.5 mL/100 g)灌胃,中毒组则用生理盐水每天(0.5 mL/100 g)灌胃。不同时间处死大鼠,用RT-PCR和免疫组化方法检测各组大鼠肝组织中Integrinα1的表达。【结果】RT-PCR和免疫组化结果显示,治疗组大鼠肝中Integrinα1 mRNA及蛋白的表达均高于中毒组,两组之间的差异具有统计学意义(P<0.05)。【结论】五味子应用于CCl4中毒大鼠急性肝损伤的治疗,可以促进肝细胞Integrinα1 mRNA及蛋白的表达,提示五味子对大鼠肝脏的保护作用可能与调节Integrinα1的代谢具有相关关系。[Objective]To investigate the dynamic expression of integrin α1 in CCl4-intoxicated rat liver, and to explore the effect and significance of schisandrin on expression of integrin α1 in CCl4-intoxicated rat hepatocytes. [Methods]The Wistar rats were randomly divided into the control group, the intoxicated group and the treated group. The rats of intoxicated and treated groups were given a single dosage(0.2mL/100g body mass) of peritoneal injection of CCl4 (200 mL/L in vegetable oil) to establish rat models of acute hepatic injury. The rats of treated group were given the extract of schisandrin (0.5 mL/100g) by intragastric administration once daily. The rats of intoxicated group were treated with normal saline (0.5 mL/100g) instead. The rats were sacrificed at different time pionts. The expression of integrin α1 in the hepatic tissue of all three groups was detected by RT-PCR and immunohistochemical method. [Results] It showed that the expression of integrin α1 mRNA and protein in rat hepatocytes of the treated group was significantly increased compared with that of the intoxicated group. The difference between the two groups was statistically significant ( P 〈0. 05). [Conclusion]Schisandrin for the treatment of acute hepatic injury induced by CCl4 can promote the expression of integrin α1 mRNA and protein in hepatocytes, which suggests that the protective effect of schisandrin on rat liver is associated with regulation of metabolism of integrin α1 in hepatic tissue.
