摘  要:目的探讨高压氧对全脑缺血大鼠模型海马神经元凋亡及代谢的影响。方法成年雄性SD大鼠30只,随机分为全脑缺血未治疗组和高压氧治疗组。采用改良的Pulsineli法制作全脑缺血大鼠模型。治疗组给予高压氧1 h/d,连续5 d。应用Nissl染色检测大鼠海马神经元数,免疫组化检测大鼠海马Caspase-3蛋白表达。结果高压氧治疗组与未治疗组相比大鼠海马CA1区神经元计数明显增加,差异有显著性(P<0.05)。高压氧治疗组大鼠海马CA1区Caspase-3蛋白表达阳性细胞计数明显低于未治疗组,差异有显著性(P<0.05)。结论严重全脑缺血损伤急性期应用高压氧可抑制凋亡相关基因的表达,改善脑组织的能量代谢,具有神经保护作用。Objective To investigate the effects of hyperbaric oxygen (HBO) on hippocampal neurons following the global cerebral ischemia/reperfusion (I/R) in rats. Methods Thirty adult male SpragueDawley rats were randomly divided into 2 groups:global ischemia-hypotension, HBO treatment after global ischemia-hypotension ( n = 15 respectively). The global I/R model was established by the modified Pulsineli 4 vessel occlusion method. After modeling, the rats in HBO treatment group were exposed to HBO for lh per day for 5d, Nissl staining was used for neuroncounting and immunohistochemical staining was employed to detect the expression of apoptosis associated Caspase-3 gene. Results The number of neurons in the hippocampal CA1 area in the HBO treatment group was significantly higher than that in the untreatment group (P 〈 0.05). The positive rate of Caspase-3 in the hippocampal CA1 area in the HBO treatment group was lower than that in untreatment group ( P 〈 0. 05 ). Conclusion HBO could decrease the expression of apoptotic genes, which reduces apoptosis after global ischemia-hypotension.
