RAM-HPLC测定兔眼房水中丝裂霉素C的含量

摘　　要：目的测定兔眼房水中缓释丝裂霉素C的含量。方法使用TSP高效液相色谱仪，Anastar工作站；分析柱：RAM（4．6mm×150mm，5μm）；流动相：甲醇．水（10：90），流速：1．0mL·min^-1，检测波长：365nm。结果色谱峰分离度良好，丝裂霉素在0．001～0．005mg·mL^-1内线性关系良好，平均回收率为99，1％，102．3％，103．6％，RSD为3．3％，2．9％，1．8％。结论本法准确简单，精密度好，是测定兔眼房水中缓释丝裂霉素C释放过程的适宜方法。OBJECTIVE To simultaneously determine the concentration of mitomycin C （MMC） in aqeous of rabbit eyes. METHODS A direct injection method for determination of MMC in body fluid and Solution by restricted-access media（RAM） -HPLC was developed. The mobile phase consisted of menthol-water（ 10： 90） at a flow rate of 1.0 mL · min^-1 and detection wavelength of 365 nm. RESULTS MMC was well separated. The assay was linear between 0. 001 - 0. 005 mg· mL^-1. The average recovery was 99.1% ,102.3% ,103.6% ,and the RSD was 3.3% ,2.9%, 1, 8%. CONCLUSION The assay method is simple,reliable,and can be used for the determination of MMC in rabbit eyes.
