摘 要:目的研究不同浓度肽聚糖(PGN)、脂多糖(LPS)、聚肌胞苷酸(Poly I:C)对过敏性支气管哮喘患者嗜碱性粒细胞释放能力的作用,进而了解过敏性支气管哮喘患者嗜碱性粒细胞上Toll样受体(TLR)的活性。方法分别应用不同质量浓度PGN、LPS、Poly I:C刺激过敏性支气管哮喘患者的外周血单个核细胞(PBMC),观察嗜碱性粒细胞脱颗粒情况,并与正常人作比较。同时检测尘螨刺激后嗜碱性粒细胞的脱颗粒程度,以分析嗜碱性粒细胞释放能力的改变与其表面IgE受体的关系。结果100 mg/L PGN刺激过敏性支气管哮喘患者嗜碱性粒细胞的脱颗粒指数为(40.73±10.88)%,显著高于其刺激正常人嗜碱性粒细胞的(15.23±8.96)%(P>0.01),亦显著高于50μg/L LPS刺激的(19.63±8.92)%和50 mg/L Poly I:C刺激的(22.50±7.89)%(P值均<0.01)。PGN刺激嗜碱性粒细胞脱颗粒指数与粉尘螨刺激脱颗粒指数不相关(P>0.05)。结论PGN可增强过敏性支气管哮喘患者嗜碱性粒细胞的释放能力,嗜碱性粒细胞上存在相应的TLR活性受体。Objective To study the effects of different concentrations of peptidoglycan (PGN), lipopolysaccharides (LPS), polyinosinic polycytidylic acid (Poly I: C) on the histamine releasing property of basophils of allergic asthma patients, so as to know about the activity of Toll-like receptor (TLR) on the basophils in allergic asthma patients. Methods Different concentrations of PGN, LPS, and Poly I:C were used to stimulate the peripheral blood mononuelear cells (PBMC) of allergic asthma patients. The basophil degranulation was observed to assess the histamine releasing property, and the results were compared with those of normal Subjects. In order to analyze the relation between IgE receptor and the changes of histamine releasing property, mite-induced basophil degranulation was also observed. Results In PGN-induced basophil degranulation index in allergic asthma patients was significantly higher than that in normal subjects ([40.73 ± 10. 883]% vs. [15.23 ± 8.96]%, P〈0.01) and that stimulated by LPS and Poly I:C ([40.73±10.88]% vs. [19.63±8.92]% , [22.50± 7.89]%, both P〈0.01). The basophil degranulation index stimulated by PGN was not related to that stimulated by mites (P〉0.05). Conclusion PGN can enhance the histamine releasing property of basophils of allergic asthma patients, and active TLR2 exists on basophils.
