摘 要:目的观察反复高热惊厥(FS)大鼠脑组织核因子-κB(NF-κB)的表达,探讨NF-κB在FS大鼠脑损伤发病中的意义。方法雄性SD大鼠51只按随机数字法分为正常对照组(n=14)、高热对照组(n=19)和FS组(n=18)。热水浴法建立FS模型;予热水浴而未出现FS为高热对照组;正常对照组未予任何处理。免疫组织化学检测NF-κB在各组大鼠脑组织各区的表达,原位末端标记法检测各组大鼠脑组织细胞凋亡。采用SPSS13.0软件进行统计学处理。结果FS组大鼠脑组织各区NF-κB阳性细胞数明显高于正常对照组和高热对照组(Pa<0.01);高热对照组大鼠脑组织各区NF-κB阳性细胞数也明显高于正常对照组(Pa<0.01)。FS组大鼠脑组织各区NF-κB阳性细胞数从高到低依次为海马齿状回[(26.10±2.40)/HP]、CA3区[(24.30±2.0)/HP]、CA1区[(22.50±3.98)/HP]、颞叶皮层[(22.10±2.33)/HP],这种部位差异与原位末端标记相一致,凋亡指数从高到低依次为海马齿状回(32.65±2.14),CA3区(28.99±1.16),CA1区(24.28±0.92),颞叶皮层(22.19±1.06)。结论反复FS致脑损伤后,NF-κB在脑组织中的表达存在部位差异,且对神经细胞的凋亡起重要作用。Objective To observe the expressions of nuclear factor - κB ( NF - κB) in brain tissue of rats with repeated febrile seizures (FS) and explore its significance in brain injury of rats with FS. Methods Fifty - one male SD rats were divided into 3 groups according to random number method: normal group (n = 14) , hyperthermic group (n = 19), FS group( n = 18 ). FS models were induced by placing rats in warm bath;the rats without FS after warm - bath were assigned as hyperthermic group ; the normal controls received no treatment. The expression of NF - κB was measured by immunohistochemistry. The apoptosis of brain tissue was determined by terminal deoxynucleotidyl transferase biotin - dUTP nick end labeling(TUNEL). SPSS 13.0 software was applied for statistical treatment. Results In FS group, the number of the NF - κB positive neuron increased much more than that of hyperthermic group and normal group(Pa 〈 0.01 ), and that in hyperthermic group was bighter than that in normal group ( Pa 〈 0. 01 ). The number of NF - κB varied in dentate gyrus(DG) [ (26.10 ± 2.40)/HP ], CA3 [ (24.30 ± 2.0)/HP1 , CA1 [ (22.50 ± 3.98 )/HP], and cortex of temporal lobe (CTL) [ (22.10 ± 2.33 )/HP]. The expression characteristics were correlated with TUNEL. Apoptotic index from high to low were in DG (32.65 ± 2.14), CA3 (28.99 ± 1. 16), and CAl(24.28 ± 0.92) , and CTL (22.19 ± 1.06). Conclusions The expression of NF - κB was different in the brain region after repeated FS and correlated with apoptosis.
