摘 要:选择合适浓度的DMSO溶剂,观察和比较糖苷和苷元对K562细胞增殖的影响.采用MTT法测定DMSO、木犀草素和木犀草苷对K562细胞增殖的影响,对结果进行了统计分析.处理结果显示:DMSO作为溶剂对K562细胞增殖产生影响的最小的浓度为0.1%.梯度实验显示:木犀草素和木犀草苷同它们的抑制作用都有量效关系.结果表明:最好选用0.1%DMSO作为观察药物影响K562细胞增殖实验的助溶剂,浓度偏大或偏小都会表现出它对K562细胞增殖的影响;在0.1%DMSO中,木犀草素和木犀草苷对K562细胞增殖均可表现出抑制作用,且呈浓度依赖性;木犀草苷的作用比木犀草素强;1.15μmol/L的木犀草苷在水溶液中的作用比在0.1%DMSO中的强,且差异极其显著.In a tested compatible concentration of dimethylsulfoxide ( DMSO), the effects of luteolin and luteolin-7-O-glucoside on proliferation of K562 cells had been compared. MTT assay was used for the tests, and results were statistically analysed. Results showed that DMSO in 0. 1% concentration had less effect on the proliferation of K562 cells than it in other concentrations, which made 0.1% DMSO the most compatible to solve aglycone and glycosides for further researches on their effects on the prolif- eration of K562 cells. Furthermore the dose - dependency on inhibitory effect was found in both of luteolin and luteolin-7-O-glucoside by testing in the concentrations of 1.15 × 10^-8mol/L, 1.15 × 10^-7mol/L and 1.15 × 10^-6mol/L. It could be concluded that 0.1% was the optimum concentration for DMSO being the menstruum. Both Luteolin and luteolin-7-O-glucoside had inhibitory effects on proliferation of K562 cells, but the effect of luteolin-7-O-glucoside might be much stronger.
