乳结消片对乳腺增生病大鼠雌二醇、孕酮水平的影响

摘　　要：目的:探讨乳结消片治疗乳腺增生病的机制。方法:采用肌内注射雌激素及孕激素的方法建立大鼠乳腺增生病理模型,造模成功后,将60只SD大鼠随机分为6组,分别为乳结消大、中、小剂量组,乳癖消组,模型对照组和正常对照组,每组10只,乳结消大、中、小剂量组按照6.0、3.0、1.5 g/kg的剂量分别灌服乳结消片,乳癖消组按照1.0 g/kg的剂量灌服乳癖消片,模型对照组和正常对照组灌服蒸馏水(20 mL/kg)。应用放射免疫法测定各组血清雌二醇和孕酮含量的变化。结果:模型对照组雌二醇含量明显高于正常对照组(P<0.05),孕酮含量明显低于正常对照组(P<0.01)。给药结束后,乳结消大、中、小剂量组和乳癖消组雌二醇含量明显低于模型对照组(P<0.05),其中乳结消大剂量组明显低于乳癖消组(P<0.05)。乳结消大、中、小剂量组和乳癖消组孕酮含量明显高于模型对照组(P<0.01)。结论:纯中药制剂乳结消片对乳腺增生病大鼠血清雌二醇和孕酮水平有调节作用,是该药治疗和预防乳腺增生病的机制之一。Objective：To explore the mechanism of Rujiexiao tablets in treating mammary gland hyperplasia. Methods： Rat model of mammary gland hyperplasia was established by intraperitoneal injection of estrogen. After model building,60 rats were divided into six groups randomly;high-dose, middle-dose,low-dose of Rujiexiao tablets group, Rupixiao tablets group, the model group and the normal group,with 10 in each group. High-dose, middle-dose, low-dose of Rujiexiao tablets group were injected with Rujiexiao tablets according to 6.0, 3.0, 1.5g/kg, Rupixiao tablets group was injected with Rupixiao tablets according to 1.0 g/kg, the model group and the normal group were injected with distilled water according to 20 mL/kg. Radioimmunoassay was used to detect the change of estradiol and progesterone hormone level in the content of rat serum. Results：The estradiol levels of model group was significantly higher than that of the normal control group （P〈0.05） and the progesterone levels was significantly lower than that of the normal control group（P〈0.01）. After the administration, the estradiol levels of high-dose,middle-dose,low-dose of Rujiexiao tablets group and Rupixiao tablets group was significantly lower than that of the model group（P〈0.05）and the high-dose of Rujiexiao tablets group was better than the Rupixiao tablets group （P〈0.05）. The progesterone levels of high-dose,middle-dose,low-dose of Rujiexiao tablets group and Rupixiao tablets group was significantly higher than that of the model group （P〈0.01）. Conclusion：Rujiexiao tablets can regulate estradiol and progesterone hormone levels in rats with mammary gland hyperpla- sia,which is one of the mechanisms of preventing and treating mammary gland hyperplasia.
