HPLC法同时测定制马钱子中4种生物碱类成分的含量

摘　　要：目的:建立HPLC法制测定马钱子中4种生物碱类成分——马钱子碱、士的宁、马钱子碱氮氧化物、士的宁氮氧化物的含量。方法:采用C18柱,流动相乙腈(A),0.2%乙酸-0.2%三乙胺(B)梯度洗脱;流量:1 mL.min-1;检测波长为254 nm;柱温:30℃;进样体积:10μL。结果:4种生物碱类成分得到良好分离,平均加样回收率:马钱子碱、士的宁、马钱子碱氮氧化物,士的宁氮氧化物分别为:100.1%、100.0%、99.8%、99.8%;RSD分别为1.17%、1.19%、1.21%、1.21%。结论:本方法结果准确,便于操作,为马钱子的质量控制提供参考。Objective： To establish a method for the determination of four alkaloids in processed Semen Strychni. Methods ：RP-HPLC was used to determing brucine, strychnine, brucine N-oxide and strychnine N-oxide. The separation was performed on C18 colunm with acetonitrile（A）, 0. 2% acetic acid water and 0. 2% triethylamine water（B） being used as a gradient program, elute rate of 1 mL·min^-1. Resuits： 4 alkaloids were separated well. Average recoveries were 100. 1% for brucine, 100. 0% for strychnine, 99. 8% for brucine N-oxide, 99. 8% for strychnine N-oxide, respectively. Concluion. The method is simple and rapid, and suitable for quality control of processed Semen Strychni.
