摘 要:文章对猪生长激素(PGH)的基因表达、包涵体变复性、猪生长激素释放因子(PGRF)等方面的研究进展进行综述.猪生长激素基因往往被克隆在一定的表达载体上,随后在大肠杆菌中表达,表达产物经纯化等处理即可获得纯品.然而猪生长激素基因在表达的过程中,往往以没有生物活性的包涵体形式存在,因而对包涵体的处理则成为影响生长激素产量的决定性因素.通过蛋白质变复性实验可以获得可溶性的且具有生物活性的蛋白质,而蛋白质变复性的效率受到许多因素的影响,包括包涵体的释放、溶解等等.猪生长激素释放因子可促进生长激素的合成与分泌,提高动物的生长速度.The gene structure and it's expression, denature and re-nature of the inclusion bodies of the porcine growth hormone (PGH) have been reviewed. To obtain purified PGH,the PGH gene is cloned to the certain expressing vector for over-expression in E. coll. Unfortunately,the PGH is found in E. coli as the form of inclusion bodies and is inactive so far. The bioactive PGH can be obtained by the means of dissolving the inclusion body of PGH in denature reagents. The denature-renature procedure of the inclusion bodies becomes the key factor to the product of PGH. During the processing of inclusion bodies, there are many factors affecting the efficiency of dissolving and re-nature,such as the releasing quantity of the inclusion bodies from the E. coli cell and dissolution of the inclusion bodies in reagent and so on. The porcine growth release factor can promote the synthesis and secretion of the porcine growth hormone to advance the growth of animals, which is becoming the researching focus of porcine growth regulator in recent year.
