摘 要:目的:观察思的明(SDM)对纳洛酮催促下的吗啡戒断大鼠不同脑区谷氨酸(Glu)含量的影响。方法:以剂量递增法连续皮下注射(sc)吗啡,建立吗啡依赖大鼠模型,取纳洛酮催促戒断后各组大鼠的皮层和海马。采用2,4-二硝基氟苯(DN-FB)柱前衍生高效液相色谱法(HPLC)测定皮层和海马中Glu的含量。结果:与生理盐水(NS)组比较,SDM对照组皮层和海马中Glu含量差异无显著性,模型组大鼠皮层和海马内Glu的含量明显增高。与模型组比较,SDM治疗组、阳性药可乐定组大鼠皮层和海马中Glu的含量均明显降低。结论:SDM能调节中枢的Glu含量,这可能与SDM可显著抑制大鼠吗啡戒断症状有关。OBJECTIVE To study the effect of Sidiming (SDM) on glutamic acid (Glu) content of the brain cortex and hip pocampus in morphine withdrawal rats induced by naloxone. METHODS A morphine dependent model of rats was established by subcutaneous injection (sc) of morphine in gradually increasing dosages. The brain cortex and hippocampus were collected from the morphine withdrawal rats induced by naloxone and the Glu content in the cortex and hippocampus was determined by HPLC with pre-column 2, 4 dinitro-fluorobenzene (DNFB) derivatization. RESULTS Compared with the NS control group, Glu content in the cortex and hipp0campus was not significantly changed in SDM control group, but it was significantly higher in the model group. Compared with the model group, Glu content in the cortex and hippocampus was significantly lower in both the SDM treatment group and the positive control clonidine group. CONCLUSION SDM can adjust the content of Glu in the cortex and hippocampus in the morphine withdrawal rats, which may be related to attenuating the morphine withdrawal symptoms.
