摘  要:目的观察血管紧张-2(Ang-2)对血管内皮细胞(VEC)内[Ca2+]i及线粒体膜电位水平的影响。方法将VEC分为空白对照组和Ang-2处理组。用激光共聚焦显微镜测量各组细胞的[Ca2+]i和线粒体膜电位。结果空白对照组的细胞内[Ca2+]i为(24.781±1.350),Ang-2组的细胞内[Ca2+]i为(47.384±1.323),Ang-2组的细胞内[Ca2+]i,明显高于空白对照组(P<0.01)。空白对照组的线粒体膜电位水平为(36.578±1.1470),Ang-2组的线粒体膜电位水平为(9.016±1.124),Ang-2组的线粒体膜电位水平明显低于空白对照组(P<0.01)。结论Ang-2可引起VEC内[Ca2+]i和线粒体膜电位水平的明显改变。Objective To explore the effects of angiotensin - 2 ( Ang - 2) on intracellular free calcium concentration and mitochondria membrane potential in vascular endothelial cell(VEC). Methods VEC were divided into control group and Ang - 2 group. Intracellular free calcium concentration and mitochondria membrane potential were measured by laser scanning confocal microscopy. Results Intracellular free calcium concentration of control group was 24. 781 ± 1.350. Intracellular free calcium concentration of Ang - 2 group was 47. 384 ± 1. 323. Intracellular free calcium concentration of Ang - 2 group was significantly higher than that of control group(P 〈0.01 ). Mitochondria membrane potential of control group was 36. 578 ± 1. 147. Mitochondria membrane potential of Ang - 2 group was 9. 016 ± 1. 124 and significantly lower than that of control group(P 〈0. 01 ). Conclusion Ang -2 can induce obvious changes of intracellular free calcium concentration and mitochondria membrane potential of VEC.
