摘  要:目的观察弓形虫可溶性速殖子抗原(soluble tachyzoite antigen,STAg)滴鼻、灌胃和皮下注射免疫小鼠诱导的抗弓形虫感染作用。方法40只6~7周龄BALB/c小鼠随机分为4组,前3组分别以20μgSTAg滴鼻、灌胃或皮下注射免疫小鼠2次,间隔2周,对照组不做处理。末次免疫后14d,用速殖子2×10^4个/只灌胃攻击所有小鼠,观察小鼠健康状况,记录体重,计算存活率。攻虫后30d颈椎脱位处死小鼠,计数脾和脑组织速殖子。结果滴鼻免疫小鼠的健康状况好于其他组,存活率(70%)高于皮下(60%)、灌胃(60%)和对照组(40%)。滴鼻组脾虫荷为369.48,皮下注射组脾虫荷为386.94,显著低于灌胃组(630.40)和对照组(664.93)(F=3.611,P〈0.05)。滴鼻组(36.45)、灌胃组(34.85)和皮下注射组(40.22)脑虫荷差异无统计学意义(F=1.999,P〉0.05),但均显著低于对照组(57.32)(F=1.999,P〈0.05)。结论20μg STAg滴鼻免疫小鼠诱导了较强的抗弓形虫感染作用,优于皮下和灌胃免疫。Objective To investigate the protective effect of soluble tachyzoite antigen(STAg) against Toxoplasma gondii in mice after intranasal,intragastric administration or subcutaneous injection with Toxoplasma gondii STAg. Methods Forty 6 to 7 weeks old BALB/c mice were randomly divided into four groups, The mice in intranasal,intragastric administration and subcutaneous injection group were immunized two times with 20μg STAg,at an interval of 2 weeks respectively and the control group was not immunized. On the day 14 after last immunization,all mice were challenged through intragastric administration with 2 × 10^4 tachyzoites of RH strain. The health of mice was observed day by day,recording the body weight and survival rate of the mice. The survivors were sacrificed on the day 30 after challenge;the tachyzoites in spleens and brains were counted. Results The health of mice in intranasal group was better than other groups, and the survival rate of intranasal group(70% ) was higher than that of subcutaneous injection( 60% ), intragastric administration (60%) and control group(40% ). The tachyzoite loads in spleen in intranasal(369.48) and subcutaneous injection(386. 94) groups were significantly lower than that of intragastric administration group ( 630. 40) and control group ( 664. 93, F = 3. 611, P 〈 0.05). The tachyzoite loads in brain in intranasal, intragastric administration and subcutaneous injection groups had no difference, but were signuficantly lower than that of control group ( F = 1. 999, P 〈 0. 05 ). Conclusion The mice immunized intranally with 20 μg STAg induced effective protection against Toxoplasma gondii, which outstripped that of intragastric administration and subcutaneous injection group.
