复方北豆根提取物胶体溶液的制备及抗人乳头瘤病毒的实验研究

摘　　要：目的探讨复方北豆根提取物胶体溶液抗人乳头瘤病毒(human papillomavirus,HPV)的分子机制,研究中药胶体溶液制备工艺流程,开发出适合女性HPV感染的新型药物释放系统。方法从北豆根中提取总碱,从蚕蛹中提取复合氨基酸,与铜绿等矿物质络合成胶体溶液,采用荧光定量PCR技术检测胶体溶液对离体尖锐湿疣中HPV-DNA复制载量。结果复方北豆根提取物胶体溶液(1 g.mL-1)及0.1倍稀释药物使HPV6/11和HPV16/18型DNA复制载量降为(1.5±1.0)copies.mL-1,与阴性对照组HPV-DNA复制载量比较,差异有统计学意义(P<0.01)。结论北豆根总碱与蚕蛹复合氨基酸、铜绿等矿物质络合成复方北豆根提取物胶体溶液可抑制HPV-DNA复制,具有明显的高活性抗HPV作用。OBJECTIVE To develop the new kind of drug-releasing system containing colloid solution of compound Menispermum dauricum extract （CMDE） for the treatment and prevention of HPV, and to explore the molecular mechanism of anti-HPV. METHODS The colloid solution containing total alkaloids of Menspermum dauricum, bombyxmoril＇s compound amino acids and minerals including Cu2 （OH）2 CO3 and borax was prepared. The fluorogenic quantitative PCR was used to detect the antiviral effect by determining the HPV-DNA amplification. RESULTS The colloid solution of CMDE （ 1 g · mL^-1 ） and 10^-1 diluted drug （0. 1 g · mL^-1） greatly inhibited the quantity of HPV-DNA with the amplification of （1.5 ± 1.0） copies · mL^-1. Compared with the negative control groups, there were signicant difference between the two groups （P 〈 0. 01 ）. CONCLUSION The colloid solution of CMDE had a great potential anti-HPV by decreasing the duplication of HPV.
